I haven't played around with it yet but 2Dtoolkit has a new UI system with tutorials and guide.

Hi,

 it depends what you do with it and the target platform. But if you plan on heavy 2d ui or game content, indeed you need to look for ngui, 2dtoolkit and the like.

and with Unity 4, things have improved on that front.
